Note 01: This construct contains also another dif site (Part:BBa_K316002) in between the the AmyE site and the Pveg promoter. This dif site is not indicated in the main page for this part, neither is included in the DNA sequence when Get selected sequence link is clicked. The dif site was discovered when the construct was sequenced using VF2 and VR primers.

Note 02:The recognition site for PmeI restriction enzyme is GTTTAAAC (and not gaaatttc as the picture in the main page suggests). This mistake is also found in the DNA sequence generated when you press the Get selected sequence link. Nevertheless the actual DNA does indeed contains the PmeI site and digestion with PmeI enzyme produces the expected DNA fragmnets.

Note 03: Also the resistance gene is Chloramphenicol casette, and not Spectinomycin gene as indicated in "Sequence and Features" table.
